In this brief review, which is focused primarily on murine systems, we outline certain important aspects of the biology of the mast cell, including their ability to produce a variety of cytokines, describe the development of a model system (the "mast cell knock-in mouse") for studying the roles of cutaneous mast cells in biological responses in vivo, and illustrate a few examples of how this approach has been used to investigate the contributions of mast cells and their cytokines to cutaneous inflammatory or immune responses.
